A PROTOCOL FOR PREPARATION FIBRIN SEALANT FROM HUMAN PLASMA

Overview

Abstract:

Fibrin sealant is an adhesive product used to stop bleeding, seal wound during and after surgery. Commerciality available fibrin sealant are made from pooled human blood which carries the risk of blood-borne infections and expensive. Autologous fibrin sealant results in reduced risks of containmination and immunological responds compared to commercial fibrin sealant. Methods: the purpose of this paper was to propose a simple technique of harvesting fibrinogen and thrombin and preparing autologous fibrin sealant from 10 whole blood samples. Results: From autologous blood, using protamin concentration of 10mg/ml, precipitated 67.42%-96.08% of fibrinogen (avarage 81.05%) present in the plasma. Thrombin was obtained through plasma combined canxi cloride 10% incubated 370C in 15 minutes.  Autologous fibrin sealant was formed in 8 minutes on avarage when mixing fibrinogen and thrombin with ratio 1:1. Conclusions: a simple process for harverting both the fibrinogen and thrombin component from small blood volume was developed. The autologous fibrin sealant can be prepared on demand with any volume of patients’ blood within a short time.
