Abstract:
Background: In recent years, tissue culture methods and staining techniques have been increasingly developed and improved, especially the G-band staining (GTG) technique that has allowed geneticists to detect structural disorders of each chromosome such as deletions, duplications, translocations, inversions, etc. couples who are diagnosed as infertile psychological and economic burdens for patients. Therefore, testing and analyzing chromosomes in these couples are very meaningful and provide genetic advice to contribute to reproductive health care in the community and society. Objective: Determine the chromosomal formula of infertile patients by Karyotype method. Methods: Culture of peripheral blood lymphocytes according to the method of Hungerford D.A; Cell harvesting method; Seabright M tape staining method; Stain slides by G-band staining according to Seabright's method. M.; Chromosome analysis and karyotyping according to ISCN standards. Results: Out of 12 couples diagnosed with infertility, 1 couple was found to have a structural disorder of chromosomes and a translocation mutation between chromosome 2 and chromosome 10. The remaining 11 couples had normal Karyotype. Conclusion: In 12 couples diagnosed with primary infertility, 1 couple had a structural disorder of chromosomes. This disorder is a translocation mutation between chromosome 2 and chromosome 10. The remaining 11 couples are infertile due to other causes. To further investigate the causes of infertility can be done by DNA analysis methods in couples with normal Karyotype.
